KinomeView® to Serine/Threonine PhosphoScan® Proteomics

Cell Signaling Technology’s KinomeView® and PhosphoScan® proteomics services are a two-step process enabling kinome-wide quantitative analysis of cellular phosphorylation for the discovery and validation of signaling targets, kinase substrates and response biomarkers. PhosphoScan services provide in-depth yet focused phosphoproteome analysis with CST’s patented Phospho-Motif Antibodies. Each of these antibodies react broadly with diverse sets of serine, threonine and tyrosine phosphorylated sequences. Our KinomeView Service provides a first step to guide the design of PhosphoScan studies by determining which branches of the kinome are responsive to the biology or chemistry under investigation. KinomeView can identify which Phospho-Motif Antibodies will yield the greatest insights when used in PhosphoScan mass spectrometry analysis.

Tyrosine PhosphoScan® Proteomics

Tyrosine PhosphoScan® Services provide the most comprehensive strategy for global phospho-tyrosine profiling. CST’s P-Tyr-1000 mAb is more broadly reactive with p-Tyr peptides than any other antibody. Employing P-Tyr-1000 for phosphopeptide enrichment in combination with LC-MS/MS analysis yields quantitative profiles of hundreds to over a thousand non-redundant phosphorylated sequences.

Tyrosine PhosphoScan proteomics is used routinely and successful for target discovery, mutant RTK screening, lead compound characterization and biomarker discovery.

AcetylScan® (Acetylation Proteomics)

Histone acetyltransferases (HATs) and histone deacetylases (HDACs) have regulator functions in numerous aspects of cell physiology. AcetylScan® is a unique and unparalleled strategy for global analysis of HDAC and HAT activity on protein acetylation. Our AcetylScan Services utilize antibodies with high affinity to acetylated-lysine (Ac-K) peptides for enrichment of acetylated peptides from protease digested cell samples, followed by LC-MS/MS analysis for quantitative profiles of many hundreds of non-redundant acetylated sequences.

AcetylScan® Services have been applied to class I, II and III HDAC studies, and have been performed on sub-cellular compartment fractions for substrate, target pathway and compound biomarker discovery.

MethylScan® (Methylation Proteomics)

Histone methylation is crucial for proper programming of the genome during development, and misregulation of the methylation machinery can lead to diseased state including cancer. MethylScan® Service is a PTMScan® Service offering that provides a unique and unparalleled strategy for global analysis of methyltransferase and demethylase activity on protein methylation. The Service utilizes antibodies with high affinity to mono-methylated arginine and mono-methylated lysine peptides for enrichment of mono-methylated peptides from protease-digested cell or tissue samples, followed by LC-MS/MS analysis for quantitative profiles of many hundreds of non-redundant mono-methylated sequences. UbiScan® (Ubiquitination Proteomics)

Ubiquitination is gaining growing attention as an important mechanism for both signal transduction and proteolytic degradation. The UbiScan® ubiquitination proteomics method is a unique and unparalleled strategy for global analysis of cellular ubiquitination. CST has developed a proprietary ubiquitin branch (“K-GG”) XP® monoclonal antibody with specificity for a di-glycine tag that is the remnant of ubiquitin left on protein substrates after trypsin digestion. The “K-GG” XP® monoclonal antibody was generated using CST’s proprietary XMT® monoclonal technology.

In UbiScan Services, we employ the K-GG antibody on trypsin digested cell samples to enrich ubiquitinated peptides, followed by LC-MS/MS analysis for quantitative profiles of hundreds to over a thousand non-redundant ubiquitinated sequences. UbiScan® Proteomic Services are applied in comparative studies to identify substrates and biomarkers of ubiquitin pathway targets and compounds.
