DELFIA®
(Perkin Elmer) Dissociation-enhanced lanthanide fluorescence immunoassay (DELFIA) is a common time resolved fluorescent (TRF) high throughput screening method routinely used to assay the phosphorylation of peptide and protein substrates in heterogeneous biochemical kinase assays.
LANCE®/HTRF®/LanthaScreen®
are a series of popular proximity assays that use time resolved fluorescence resonance energy transfer to measure phosphorylation of a specific peptide substrate. Each assay uses a conjugated antibody donor and a unique acceptor molecule. When donor and acceptor are brought into close proximity via a biomolecular interaction, an energy transfer occurs that results in increased fluorescence of the acceptor molecule.
| Comparison Table | LANCE® | HTRF® | LanthaScreen® |
|---|---|---|---|
| Fluorescence (nm) | 665 | 665 | 520:495 ratio |
| Donor | europium chelate | europium cryptate | terbium chelate |
| Acceptor | allophycocyanin | XL665 | fluorescein |
AlphaScreen®
(Perkin Elmer) is a screening technology based on the transfer of energy from donor to acceptor microbeads brought together by a biomolecular interaction. Photosensitizers in the donor beads convert ambient oxygen to singlet state oxygen, which can induce a cascade of chemiluminescence in the acceptor bead if brought within close proximity. The emission of an AlphaScreen® signal can be measured at 520-620 nm.
