Bio-Plex nine-plex phosphoprotein assay. Following treatment of HeLa cells with TNF-alpha or IFN, the phosphorylation status of GSK3-alpha/ beta (Ser21/9), Akt (Ser473), MEK2 (Ser221), ATF-2 (Thr69), Stat3 (Tyr705), JNK (Thr183/Tyr185), p38MAPK (Thr180/Tyr182), IkB-alpha (Ser32/36), and p44/42 (Thr202/Tyr204) were assessed using a cocktail of bead-conjugated capture antibodies and biotinylated detection antibodies. With the addition of streptavidin-PE, mean fluorescence intensity (MFI) for each bead is quantified in proportion to the levels of each phosphorylated protein.

Cell Signaling Technology and Bio-Rad have partnered to develop multiplex phosphoprotein assays on Bio-Rad’s Bio-Plex® Suspension Array System. CST is developing highly specific antibodies for exclusive use in Bio-Plex phosphoprotein assays. The Bio-Plex platform enables profiling of multiple signaling proteins in a single well of a 96-well plate. Singleplex and multiplex total- and phosphoprotein assays are available from Bio-Rad as off-the-shelf and customizable x-Plex assays (see www.bio-rad.com/products/phosphoproteins for details).
