Overview of HTRF technology.
HTRF (Cisbio international) is a homogeneous time resolved fluorescence assay based on the proximity of a europium cryptate donor label and an XL665 acceptor. Proximity of cryptate-labeled phosphorylation specific antibody and streptavidin-XL665 fluorophore causes fluorescence resonance energy transfer (FRET), measured at 665nm. This value is proportional to the amount of biotinylated phosphopeptide present. Europium cryptates are ideal detection chemistries due to their inherent stability and low background fluorescence.
CST Antibodies Validated By Cisbio
Cisbio and Cell Signaling Technology have come together to offer high quality anti-phospho serine/threonine antibodies labeled with Eu3+Cryptate, the HTRF donor fluorophore. This line of labeled CST antibodies ranges from target to phospho-motif specific antibodies (recognize a single phospho-residue) to motif specific (recognize close epitopic sequence found in group of substrates). In addition, the phospho-threonine antibodies bind phosphorylated threonine residues largely independently of the neighboring amino acid sequence.
