Specificity / Sensitivity

Myc-Tag (9B11) Mouse mAb (Alexa Fluor® 488 Conjugate) detects transfected proteins containing the Myc epitope tag. The antibody recognizes the Myc-tag fused to either the amino or carboxy terminus of targeted proteins in transfected mammalian cells.

Source / Purification

Monoclonal antibody is produced by immunizing mice with a synthetic peptide (KLH-coupled) corresponding to residues 410-419 of human c-Myc (EQKLISEEDL). The antibody was conjugated to Alexa Fluor® 488 under optimal conditions with an F/P ratio of 2-5.

Flow Cytometry

Flow Cytometry

Flow cytometric analysis of COS cells, untransfected (blue) or transfected with Myc-tagged Akt (green), using Myc-Tag (9B11) Mouse mAb (Alexa Fluor® 488 Conjugate).

IF-IC

IF-IC

Confocal immunofluorescent analysis of parental COS cells (left) and COS cells expressing a carboxy-terminal Myc-tagged protein (right), using Myc-Tag (9B11) Mouse mAb (Alexa Fluor® 488 Conjugate). Blue pseudocolor = DRAQ5™ (fluorescent DNA dye).

Description

Cell Signaling Technology Antibody was conjugated to Alexa Fluor ® 488 fluorescent dye and tested in-house for direct flow cytometry and immunofuorescence in cells transfected with Myc-tagged protein.

Background

Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation and immunostaining techniques. Due to their small size, they are unlikely to affect the tagged protein's biochemical properties.

The Myc epitope tag is widely used to detect expression of recombinant proteins in bacteria, yeast, insect and mammalian cell systems.
