Protocols
- 2475:
- IHC / Paraffin, Western Blotting
Specificity / Sensitivity
PSA/KLK3 (D11E1) XP® Rabbit mAb recognizes endogenous levels of total PSA/KLK3 protein.
Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val72 of human PSA/KLK3 protein.
Western Blotting
Western blot analysis of extracts from human sperm and LNCaP and DU 145 cells using PSA/KLK3 (D11E1) XP® Rabbit mAb. DU 145 cells are negative as expected.
IHC-P (paraffin)
Immunohistochemical analysis of paraffin-embedded cell pellets, LNCaP (left) or DU 145 (right), using PSA/KLK3 (D11E1) XP® Rabbit mAb.
IHC-P (paraffin)
Immunohistochemical analysis of paraffin-embedded human prostate adenocarcinoma using PSA/KLK3 (D11E1) XP® Rabbit mAb in the presence of control peptide (left) or antigen-specific peptide (right).
Background
Kallikrein 3 (KLK3), also known as Prostate Specific Antigen (PSA), is a member of the glandular kallikrein subfamily of serine proteases (1). It is produced by prostate epithelial cells and is secreted into prostatic ducts. Upon cleavage of 7 N-terminal amino acids (2), it is activated to liquefy semen in the seminal coagulum. Although PSA/KLK3 is produced in healthy individuals, investigators have found abnormally high levels in the blood of men with advanced prostate cancer (3,4).
