Specificity / Sensitivity
Notch1 (6A5) Rabbit mAb detects endogenous levels of the full-length and the cytosolic domain of notch1 cleaved between Gly1743 and Val1744.
Source / Purification
Monoclonal antibody is produced by immunizing rabbits with a synthetic peptide (KLH-coupled) derived from the sequence of human Notch1.
Western Blotting
Western blot analysis of extracts from COS-7 cells, transfected with Notch1 (lane 1), mock transfected (lane 2) or 3T3-L1 cells differentiated for 10 days (lane 3), using Notch1 (6A5) Rabbit mAb (left) and Cleaved Notch 1 (Val1744) Antibody (2421) (right).
Background
Notch1 is a transmembrane protein functioning in development and the determination of cell-fate (1). During maturation, the notch molecule is cleaved by a furin-like convertase at its extracellular domain (2). Upon binding to a ligand such as Delta1, or upon extracellular calcium depletion, the carboxy-terminal notch1 fragment is released and further cleaved between Gly1743 and Val1744 (3,4). The resulting activated cytosolic fragment translocates to the nucleus where it activates transcription.
