Specificity / Sensitivity

PKM1/2 Antibody detects endogenous levels of total PKM (including M1 and M2) protein.

Source / Purification

Polyclonal antibodies are produced by immunizing rabbits with a synthetic peptide (KLH-coupled) derived from the sequence of human PKM. Antibodies are purified by protein A and peptide affinity chromatography.

Background

Pyruvate kinase, a glycolytic enzyme, catalyses the conversion of phosphoenolpyruvate to pyruvate. In mammals, four different isozymes of this kinase have been identified: M1, M2, L and R. M1 is the major form in brain and muscle, and M2 is found in early stage fetal cells and most adult tissues. L and R are present in the liver and red blood cells, respectively. Tumor cells usually overexpress M2 isozyme (PKM2), leading to an increase of the amount of glycolytic phospho-metabolites in these cells (1). This isozyme can exist in a tetrameric form with a high substrate affinity or a dimeric form with a low affinity for its substrate (2). In tumor cells, the dimeric form is generally predominant (2). Studies suggest that this isozyme could potentially serve as a marker for gastric and colorectal tumors (1,2)
