Background

Leucine carboxyl methyltransferase-1 (LCMT1) is an important mammalian methyltransferase that methylates the catalytic C subunit of protein phosphatase 2A (PP2A) at Leu309 (1,2). X-ray crystallography analysis reveals that the structural integrity of human LCMT1 and cofactor S-adenosylmethionine complex enables proper interaction with PP2A (3). Evidence suggests that functional LCMT1 is crucial for the ability of PP2A to form heterotrimers with the B/B55 and Bα family of regulatory subunits (4,5). Impaired LCMT1 activity is shown to disrupt mitosis and cell survival (5). In neurodegenerative models, increases in endogenous levels of LCMT1, methylated PP2A (C), and Bα induce the development and differentiation of neuroblastoma cells (5-7).
