Specificity / Sensitivity

CHD3 Antibody, CHD4 Antibody, and MTA1 (D40D1) XP® Rabbit mAb recognize endogenous levels of respective total proteins. MBD3 Antibody recognizes endogenous levels of all three isoforms of the MBD3 protein (MBD3A, MBD3B, MBD3C) and does not cross-react with MBD2. RBAP46 (V415) Antibody recognizes endogenous levels of total RBAP46 protein and does not cross-react with the RBAP48 protein. HDAC1 (10E2) Mouse mAb and HDAC2 (3F3) Mouse mAb recognize endogenous levels of respective total proteins and do not cross-react with other HDAC proteins.

Description

NuRD Complex Antibody Sampler Kit offers an economical means of detecting each target protein that composes the nucleosome remodeling and deacetylation complex (NuRD). The kit contains enough primary antibody to perform four western blot experiments with each primary antibody.

Source / Purification

Monoclonal antibodies are produced by immunizing animals with synthetic peptides corresponding to the carboxy terminus of human HDAC2 protein, the carboxy terminus of human HDAC1 protein, or residues near the carboxy terminus of human MTA1 protein. Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to human MBD3 protein, human CHD4 protein, human CHD3 protein, or residues surrounding Val415 of human RBAP46 protein. Polyclonal antibodies are purified by protein A and peptide affinity chromatography.

Background

The transcriptional repressor NuRD (nucleosome remodeling and histone deacetylase) complex is composed of multiple subunits, which include CHD3 (also known as Mi2-α), CHD4 (also known as Mi2-β), HDAC1, HDAC2, RBAP48, RBAP46, MTA1, MTA2, MTA3, and MBD3 (1-6). Both CHD3 and CHD4 contain two PHD (plant homeodomain) zinc finger domains that bind directly to HDAC1 and HDAC2. HDAC1 and HDAC2 are highly homologous and are involved in histone deacetylation, chromatin remodeling and transcriptional repression, and are also found together in the mSin3A and MeCP1 complexes (7-9). RBAP46 and RBAP48 bind to the histone fold region of histone H4 and are believed to target the NuRD complex to its histone substrates (10). MBD3, a member of the methyl-CpG-binding domain protein family, and the metastasis associated genes MTA1, MTA2, and MTA3 were all found to be integral components of the NuRD complex (2,3,7,11).
