Specificity / Sensitivity
Phospho-AMPA Receptor (GluR 1) (Ser845) (D10G5) Rabbit mAb recognizes endogenous levels of AMPA Receptor (GluR 1) protein only when phosphorylated at Ser845. While the literature refers to this residue as Ser845, it is Ser863 in the UniProt sequence P42261. Phospho-AMPA Receptor (GluR 2) (Tyr869/Tyr873/Tyr876) Antibody detects endogenous levels of GluR 2 protein only when phosphorylated at Tyr869, Tyr873, or Tyr876. It may also detect GluR 3 when phosphorylated at the conserved Tyr880, Tyr884, or Tyr887. These residues are not conserved in GluR 1 or GluR 4. AMPA Receptor (GluR 2) (D39F2) Rabbit mAb detects endogenous levels of total GluR 2 protein. The antibody is not predicted to recognize other AMPA receptor subunits (e.g. GluR 1, GluR 3, or GluR 4) based on sequence homology of the antigen. AMPA Receptor (GluR 3) (D47E3) Rabbit mAb detects endogenous levels of total GluR 3 protein. The antibody is not predicted to detect other AMPA receptor subunits (e.g. GluR 1, GluR 2, or GluR 4) based on sequence homology of the antigen. AMPA Receptor (GluR 4) (D41A11) XP® Rabbit mAb detects endogenous levels of total GluR 4 protein. AMPA Receptor (GluR 2/3/4) Antibody detects endogenous levels of total GluR 2/3/4 protein. It may also detect GluR 1.

Description
The AMPA Receptor Antibody Sampler Kit provides an economical means of evaluating the four subunits of AMPARs. The kit contains enough primary and secondary antibodies to perform four western blot experiments with each antibody.
Source / Purification
Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ser720 of human GluR 2 protein or a synthetic phosphopeptide corresponding to residues surrounding Tyr869, Tyr873, and Tyr876 of human GluR 2 protein. Polyclonal antibodies are purified by protein A and peptide affinity chromatography.Monoclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues near the amino terminus of human GluR 2 protein, surrounding Pro590 of human GluR 3 protein, surrounding Gln890 of human GluR 4 protein, or surrounding Ser845 of human AMPA Receptor (GluR 1) protein.
Background
AMPA- (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid), kainite-, and NMDA- (N-methyl-D-aspartate) receptors are the three main families of ionotropic glutamate-gated ion channels. AMPA receptors (AMPARs) are comprised of four subunits (GluR 1-4), which assemble as homo- or hetero-tetramers to mediate the majority of fast excitatory transmissions in the CNS. AMPARs are implicated in synapse formation, stabilization, and plasticity (1). AMPARs that lack GluR 2 are permeable to calcium, in contrast to GluR 2-containing AMPARs (2). Post-transcriptional modifications (alternative splicing, nuclear RNA editing) and post-translational modifications (glycosylation, phosphorylation) result in a very large number of permutations, fine-tuning the kinetic properties of AMPARs. Research studies have implicated activity changes in AMPARs in a variety of diseases including Alzheimer’s, amyotrophic lateral sclerosis (ALS), stroke, and epilepsy (1).
