Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues around Gly64 of human PIP5K1A. Antibodies are purified by protein A and peptide affinity chromatography.

Background

Phosphatidylinositol-4-phosphate 5-kinases (PIP5K) synthesize phosphatidylinositol-4,5-bisphosphate (PtdIns(4,5)P2), a key precursor in phosphoinositide signaling that also regulates some proteins and cellular processes directly. There are two subfamilies of PIP5Ks, type I and II, that generate PtdIns(4,5)P2 from distinct substrate pools. PIP5K1s use PtdIns4P as a substrate, whereas PIP5K2s use PtdIns5P (1,2). In mammalian cells, three isoforms of each PIP5K1 and PIP5K2 subfamily, encoded by distinct genes, have been characterized and named α, β and γ (3-7), and additional PIPKs likely remain to be discovered. All PIP5K isoforms are stimulated by phosphatidic acid, are extensively regulated by ARF and Rho GTPases, and inhibited by protein kinase A and PI-stimulated autophosphorylation (8).
