Specificity / Sensitivity
Each antibody in the Rag Protein Antibody Sampler Kit detects endogenous levels of its target protein. RagA (D8B5) Rabbit mAb may also detect endogenous levels of total RagB protein.
Western blot analysis of extracts from various cell lines, using Raptor (24C12) Rabbit mAb #2280.
Western blot analysis of extracts from 293, A-431, COS-7, C6, and C2C12 cells, using mTOR (7C10) Rabbit mAb #2983.
Western blot analysis of extracts from various cell types using RagB Antibody #3830.
Western blot analysis of extracts from mIMCD3, KNRK and COS-7 cells using RagA (D8B5) Rabbit mAb #4357.
Western blot analysis of extracts from various cell lines using RagC (D31G9) XP® Rabbit mAb #5466.
The Rag Protein Antibody Sampler Kit provides an economical means to evaluate the regulation of mTORC1 signaling by such proteins as Raptor, RagA, RagB, RagC, and RagD. The kit contains enough primary and secondary antibodies to perform four Western blot experiments per primary antibody.
Source / Purification
Polyclonal antibodies are produced by immunizing animals with synthetic phosphopeptides corresponding to residues in the human sequences for RagB, and RagD proteins, andcorresponding to residues near the amino terminus of human RagC protein. Polyclonal antibodies are purified by protein A and peptide affinity chromatography. Monoclonal antibodies are produced by immunizing animals with synthetic peptides corresponding to residues of human mTOR, RagA, and Raptor.
The mTORC1 kinase complex is a critical regulator of cell growth (1,2). Its activity is modulated by energy levels, growth factors, and amino acids via signaling through Akt, MAPK, and AMPK pathways (3,4). Recent studies found that the four related GTPases, RagA, RagB, RagC, and RagD, interact with raptor within the mTORC1 complex (1,2). These interactions are both necessary and sufficient for mTORC1 activation in response to amino acid signals (1,2).
