About PhosphoScan®
Advanced Proteomics for Target and Biomarker Discovery
PhosphoScan® is a uniquely powerful and patented technology that enables researchers to identify and quantify hundreds to thousands of phosphorylated sequences in cell and tissue samples and to discover phosphorylation markers of kinase-mediated pathway activation and inhibition. PhosphoScan employs phospho-residue (Tyr, Ser, Thr) motif antibodies for phosphopeptide immunoaffinity purification from cell extracts combined with LC tandem mass spectrometry to identify and quantify changes in phosphorylation levels.
Scientists at Cell Signaling Technology (CST) have used PhosphoScan to determine broad cellular PhosphoSignatures™ (phosphorylation profiles) from numerous sample types, including over 300 cultured cell lines, xenografts, mouse brain, heart, liver and lung, as well as primary human tumors. CST’s discovered PhosphoSignatures represent well over 10,000 novel sites of tyrosine phosphorylation, as well as a rapidly growing database of serine/threonine PhosphoSignatures. The tyrosine PhosphoSignatures alone represent nearly four-times the number of sites described in the scientific literature and nearly 70% of the sites discovered by CST were previously unknown based on site annotations in PhosphoSitePlus, CST’s bioinformatics resource.

The PhosphoScan® – PhosphoSitePlus™ Connection
PhosphoScan has led to many discoveries and has resulted in the identification of thousands of unpublished phosphorylation sites. Many of Cell Signaling Technology’s PhosphoScan research results are available to researchers through PhosphoSitePlus, (www.phosphosite.org) an expert-curated knowledgebase of protein phosphorylation in vertebrates.
