Background: The zyxin family of proteins includes LIMD1, ajuba, trip6 and zyxin, each of which contains three LIM domains at the carboxy-terminus. Zyxin family members associate with the actin cytoskeleton and are components of both the cell-cell junction adhesive complex and the integrin-mediated adhesive complex. They shuttle in and out of the nucleus where they may function in transcriptional activation (1).Zyxin is involved in the regulation of mechanical force-induced actin polymerization at focal adhesions (2), and in regulation of adhesion and migration, possibly through recruitment of Ena/VASP proteins to focal adhesions (3). Zyxin interacts with and may regulate the function of the tumor suppressor myopodin, which inhibits tumor growth and metastasis (4).

The PathScan® Phospho-Src (Tyr416) Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of phospho-Src (Tyr416). A phospho-Src rabbit antibody has been coated onto the microwells. After incubation with cell lysates, phospho-Src (Tyr416) is captured by the coated antibody. Following extensive washing, a Src mouse detection antibody is added to detect the captured phospho-Src (Tyr416). Anti-mouse, HRP-linked antibody is then used to recognize the bound detection antibody. The HRP substrate, TMB, is added to develop color. The magnitude of the absorbance for this developed color is proportional to the quantity of phospho-Src (Tyr416).Antibodies in kit are custom formulations specific to kit.

Src activity is regulated by tyrosine phosphorylation at two sites, but with opposing effects. While phosphorylation at Tyr416 in the activation loop of the kinase domain upregulates enzyme activity, phosphorylation at Tyr527 in the carboxy-terminal tail by Csk renders the enzyme less active (2).

Capture and detection antibodies (100X stocks) and an HRP-conjugated secondary antibody (1000X stock) are supplied. Sufficient reagents are supplied for 4 x 96 well ELISAs. The phospho-Src (Tyr416) rabbit capture antibody is coated onto a 96 well microplate overnight in PBS. After blocking, cell lysates are added followed by a total Src mouse detection antibody and anti-mouse IgG, HRP-linked antibody. HRP substrate (TMB) is then added for color development. Background: The dynamic polymerization and depolymerization of actin filaments, a process governed by external and internal signaling events, is vital for cell motility (immune cell function, migration, invasion, metastasis, angiogenesis), cell division and adhesion. Among the many regulators of actin dynamics are profilins. Profilins are conserved actin binding proteins that affect the rate of actin polymerization by binding actin monomers and promoting the exchange of ADP for ATP (reviewed in 1). Profilins bind to proteins involved in the regulation of actin dynamics including palladin (2), dynamin-1 (3), VASP (4) and N-WASP (5). In mice, knockout of the ubiquitously expressed profilin-1 indicates that the protein is essential for embryonic development (6). Profilin-2 is primarily expressed in brain and functions in the regulation of neurite outgrowth (7), membrane trafficking and endocytosis (3). The recently cloned profilin-3 is expressed in kidney and testes (8).
