Background: α-ketoglutarate dehydrogenase complex is a rate-regulating enzyme in the Krebs Cycle (1). Dihydrolipoamide succinyltransferase (DLST) is a key subunit in this complex (2). Reduction of DLST increases reactive oxygen species production, suggesting its role in oxidative stress (2). Research has shown that deficiency of DLST in mice is linked to increased oxidative stress in mitochondria, a process that may be involved in the pathogenesis of Alzheimer's disease (2).

Background: 2-oxoglutarate dehydrogenase (OGDH) is one of three enzymes in the α ketoglutarate dehydrogenase complex (OGDC) that is responsible for catalyzing a rate-regulating step of the tricarboxylic acid (Krebs) cycle. Together with dihydrolipoamide S-succinyltransferase (DLST) and dihydrolipoamide dehydrogenase (DLD), OGDH helps to convert 2-oxoglutarate to succinyl-CoA and CO2 within eukaryotic mitochondria (1). Regulation of this enzyme complex is important for mitochondrial energy metabolism within cells (2). Research studies indicate that OGDH activity within the mitochondrial matrix is regulated by multiple factors, including calcium, the adenine nucleotides ATP and ADP, and NADH (2).

This Cell Signaling Technology (CST) antibody is conjugated to Alexa Fluor® 488 fluorescent dye and tested in-house for direct flow cytometry in human cells. CST expects that Phospho-Stat5 (Tyr694) (C71E5) Rabbit mAb (Alexa Fluor® 488) will display the same species cross-reactivity as the unconjugated antibody (Phospho-Stat5 (Tyr694) (C71E5) Rabbit mAb #9314).

Background: Stat5 is activated in response to a wide variety of ligands including IL-2, GM-CSF, growth hormone and prolactin. Phosphorylation at Tyr694 is obligatory for Stat5 activation (1,2). This phosphorylation is mediated by Src upon erythropoietin stimulation (3). Stat5 is constitutively active in some leukemic cell types (4). Phosphorylated Stat5 is found in some endothelial cells treated with IL-3, which suggests its involvement in angiogenesis and cell motility (5). Stat5a and Stat5b are independently regulated and activated in various cell types. For instance, interferon treatment predominantly activates Stat5a in U-937 cells and Stat5b in HeLa cells (6).

SignalSilence® Stat5 siRNA I from Cell Signaling Technology (CST) allows the researcher to specifically inhibit Stat5 expression using RNA interference, a method whereby gene expression can be selectively silenced through the delivery of double stranded RNA molecules into the cell. All SignalSilence® siRNA products from CST are rigorously tested in-house and have been shown to reduce target protein expression by western analysis.

Background: Glutamate oxaloacetate transaminase 1 (GOT1) catalyzes the interconversion of aspartate and oxaloacetate (1). The increased transamination primarily catalyzed by GOT1 leads to elevated levels of 2-hydroxyglutarate, which promotes methylation of the Foxp3 gene locus, inhibits Foxp3 expression and activates T helper 17 (TH17) cell differentiation (2). In addition, GOT1 is critical to the survival of cells with electron transport chain inhibition by generating aspartate, a metabolite determining the proliferation of these cells (3-4). Studies also show that GOT1 plays a key role in the noncanonical glutamine pathway that supports liver tumorigenesis (5).

CST’s PathScan® Phospho-Stat5 (Tyr694) Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of Stat5a when phosphorylated at Tyr694 and Stat5b at Tyr699. A Phospho-Stat5 Mouse Antibody has been coated onto the microwells. After incubation with cell lysates, phospho-Stat5a (Tyr694) and phospho-Stat5b (Tyr699) proteins are captured by the coated antibody. Following extensive washing, a Stat5 Rabbit Detection Antibody is added to detect the captured phospho-Stat5a and phospho-Stat5b protein. Anti-rabbit IgG, HRP-linked antibody is then used to recognize the bound detection antibody. HRP substrate, TMB, is added to develop color. The magnitude of absorbance for this developed color is proportional to the quantity of Stat5a phosphorylated at Tyr694 and Stat5b phosphorylated at Tyr699.Antibodies in kit are custom formulations specific to kit.
