Background: CD2 is a transmembrane glycoprotein expressed early in thymocyte development and present on most circulating T cells (1, 2). CD2 plays a role in T cell adhesion through binding to its ligand CD58 (LFA-3) (3). Stimulation of CD2 also leads to T cell activation and proliferation (2). T cells from mice deficient in both CD2 and CD28 have severe defects in T cell activation and function, while T cells deficient in either CD2 or CD28 are still capable of mounting a response, suggesting that CD2 and CD28 may have overlapping functions and may be able to compensate for each other (4). In addition, engagement of CD2 and CD58 was recently demonstrated to be the primary costimulatory signal in T cells that lack CD28 (5). CD2 expression also distinguishes a subset of plasmacytoid dendritic cells found in tumors and tonsils that express lysozyme, higher levels of IL-12 p40, and higher levels of CD80 (6).

Background: The Thy1/CD90 cell surface antigen is a GPI-anchored, developmentally regulated protein involved in signaling cascades that mediate neurite outgrowth, T cell activation, tumor suppression, apoptosis, and fibrosis (1). Thy1/CD90 is highly expressed on the surface of adult neurons and is thought to play a role in modulating adhesive and migratory events, such as neurite extension (1,2). Decreased Thy1/CD90 mRNA and protein expression is associated with the development of epithelial ovarian cancer, suggesting a role as a putative tumor suppressor gene of human ovarian cancer (3,4). Research studies indicate that Thy1/CD90 knockout mice have impaired cutaneous immune responses and abnormal retinal development (5,6). Thy1/CD90 is epigenetically regulated or deregulated in some disease states, such as pulmonary fibrosis. The potentially reversible hypermethylation of the Thy1/CD90 promoter offers the possibility of novel therapeutic options in this disease (7).

Background: CD59 is a GPI-anchored membrane protein that functions as inhibitor of the complement membrane attack complex (MAC). CD59 binds to complement components C8 and C9, preventing C9 polymerization and insertion into membranes, therefore inhibiting the complement-dependent cytolysis (CDC) (1). CD59 is a ubiquitously expressed cell membrane protein that protects cells from CDC. Rare cases of CD59 deficiency have been reported to cause paroxysmal nocturnal hemoglobinuria in human patients (2,3). Expression of CD59 on tumor cells and viral infected cells makes them resist antibody-dependent complement-mediated lysis. Potent inhibitors for CD59 have been actively pursued for therapeutic applications (4,5). In addition, CD59 may regulate insulin secretion by modulating exocytosis (6).
