It is possible that longer treatments (e.g., 24 hours) can lead to diminished signals for phospho-Smad2 and phospho-Smad3 because of Smad7 induction.

It is possible to see a doublet with our Phospho-c-Jun (Ser73) (D47G9) XP Rabbit mAb #3270 in some extracts. This is because #3230 can recognize JunD phosphorylated at Ser100. Phospho-c-Jun (Ser73) is the higher band at approximately 48 kDa, whereas phospho-JunD (Ser100) is the lower band at approximately 42 kDa.

Antibodies directed against phosphorylated tyrosine (PY) represent a significant challenge with respect to IHC assays. It is not uncommon for anti-PY antibodies to demonstrate cross-reactivity with other proteins with phosphorylated tyrosines. This is common with RTKs for which membrane staining is typically seen. So, if one is working with an antibody directed against one phospho-RTK and there is some cross-reactivity with other phospho-RTKs, one cannot safely interpret a signal at the membrane to be specific.

If CST recommended buffers are being used for western blotting and high background or additional bands persist when probing for phospho-AMPK (Thr172), the issue could be related to the lysate sample. In general, tissue samples can be prone to high background by western blotting when compared to cultured cell lines, and liver tissue samples (a high expressing model for AMPK) in particular tend to show high background in the form of multiple bands when probing for phospho-AMPK. Sonication and the use of proper buffers for western blotting can help to alleviate this issue in some instances. Our Phospho-IGF-I Receptor β (Tyr1135/1136)/Insulin Receptor β (Tyr1150/1151) (19H7) Rabbit mAb #3024 recognizes Tyr1189/1190 in isoform 1 (Long) and Tyr1177/1178 in isoform 2 (Short) of human insulin receptor β (see Uniprot #P06213; https://www.uniprot.org/uniprotkb/P06213/entry#sequences). “Tyr1150/1151” reflects the position of the phosphosites in isoform 2 when the 27 amino acid signal peptide is removed from the N-terminus.

It is critical that our Phospho-p53 (Ser15) Antibody #9284 be diluted into 5% w/v non-fat dry milk, 1X TBS, 0.1% Tween^® 20 and incubated at 4°C with gentle shaking, overnight. Diluting the antibody in BSA will result in high background on the blot.
