The CD44 gene encodes 20 exons. Alternative splicing gives rise to CD44s and multiple CD44 variants (CD44v). CD44s is encoded by constant exons 1-5, 16-18, and 20. The CD44 variants are produced by the insertion of variant exons 6–15, typically identified as v1–v10. The CD44 variants can contain one or more variant regions, such as CD44v3, CD44v6, CD44v4–10, or CD44v8–10. Exon 19 is spliced out in all forms of CD44.
Our CD44 (156-3C11) Mouse mAb #3570 recognizes residues surrounding Pro210 of human CD44 (UniProt ID #P16070; https://www.uniprot.org/uniprot/P16070), which are encoded by exon 5. Therefore, #3570 is predicted to recognize all variants of human CD44.

We do not observe any bias toward euchromatin or heterochromatin in our assays. In CUT&RUN, the target-specific antibody recruits the pAG-MNase and directs digestion to chromatin that is directly adjacent to the antibody, whether that be in euchromatin or heterochromatin. In this case, the active tethering of pAG-MNase to the chromatin allows for digestion to occur even in less accessible heterochromatin. Bio-Techne has tested various commercial buffers and common buffer components for compatibility with Simple Western™ and has found that some buffer components may negatively affect Simple Western™ results. For reference, Bio-Techne provides a helpful buffer compatibility chart on their website. At CST we use samples prepared in either SDS sample buffer (1X) (Blue Loading Buffer Pack #7722 or Red Loading Buffer Pack #7723) or Cell Lysis Buffer (1X) #9803. These buffers did not impact run performance in-house.
