How to Sonicate Chromatin: Enzymatic ChIP Protocol

 

Chromatin fragmentation can be performed using either sonication or enzymatic digestion. Despite its name, the latter still requires a brief sonication step to break open nuclear membranes and release the chromatin after digestion with micrococcal nuclease.
