Product Usage Information

Biotinylated antibodies are ideal for immunoassay technologies and high-throughput ELISA platforms that require antibody pairs where both antibodies are from the same host. Platforms utilizing biotinylated antibodies include, but are not limited to, MSD, xMAP, Quanterix Simoa, AlphaLISA, AlphaScreen, HTRF, LANCE, and TR-FRET.

Optimal dilutions/working concentrations should be determined by the end user. Please contact us if you require the antibody clone biotinylated at a different concentration, a carrier-free formulation, or a more customized packaging solution.

Storage

Supplied in 140 mM NaCl, 3 mM KCl, 10 mM sodium phosphate (pH 7.4) dibasic, 2 mM potassium phosphate monobasic, 2 mg/mL BSA, and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

IRF-5 (E9I4Z) Rabbit Monoclonal Antibody (Biotinylated) recognizes human IRF-5 protein and is also reactive with mouse IRF-5 protein; however, this antibody conjugate is not suggested for immunofluorescent analysis of human samples. Instead, IRF-5 (E7F9W) Rabbit Monoclonal Antibody #76983 is recommended for immunofluorescent analysis of human samples.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human IRF-5 protein.

Description

This Cell Signaling Technology® antibody is the biotinylated version of the unconjugated IRF-5 (E9I4Z) Rabbit mAb #96527 and is expected to exhibit the same species cross-reactivity. The concentration of the biotinylated antibody is 500 μg/mL. Peptide ELISA data were generated using the biotinylated antibody.

Background

Interferon regulatory factors (IRFs) comprise a family of transcription factors that function within the Jak/Stat pathway to regulate interferon (IFN) and IFN-inducible gene expression in response to viral infection (1). IRFs play an important role in pathogen defense, autoimmunity, lymphocyte development, cell growth, and susceptibility to transformation. The IRF family includes nine members: IRF-1, IRF-2, IRF-9/ISGF3γ, IRF-3, IRF-4 (Pip/LSIRF/ICSAT), IRF-5, IRF-6, IRF-7, and IRF-8/ICSBP. All IRF proteins share homology in their amino-terminal DNA-binding domains. IRF family members regulate transcription through interactions with proteins that share similar DNA-binding motifs, such as IFN-stimulated response elements (ISRE), IFN consensus sequences (ICS), and IFN regulatory elements (IRF-E) (2).

Peptide ELISA was performed on a plate coated with a synthetic peptide corresponding to residues near the carboxy terminus of human IRF-5 protein. The relationship between the concentration of IRF-5 (E9I4Z) Rabbit mAb biotinylated (blue) and non-biotinylated (red) and the absorbance at 450 nm using a peptide-coated plate is shown. Streptavidin-HRP #3999, TMB Substrate #7004, and STOP Solution #7002 were used for color development.
