Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to a fragment of the extracellular domain of human PTPRJ/DEP-1 protein.

Background

Protein-tyrosine phosphatase receptor type J (PTPRJ), also known as density-enhanced phosphatase-1 (DEP-1), is a heavily glycosylated, type 1 transmembrane protein. The extracellular region of PTPRJ/DEP-1 contains multiple copies of the fibronectin type III motif, while its intracellular region harbors a single PTP catalytic domain (1). PTPRJ/DEP-1 is expressed at varying levels on multiple hematopoietic cell lineages and has been shown to modulate signaling downstream of the TCR complex (2,3). Research studies have suggested that PTPRJ/DEP-1 also plays a role in non-hematopoietic cell biology through its ability to inhibit cell growth in response to increasing cell density (4). Indeed, PTPRJ/DEP-1-dependent dephosphorylation of mitogenic molecules, such as c-Met and PDGFR-β, has been shown to attenuate signaling downstream of these RTKs (5,6). In many contexts, PTPRJ/DEP-1 functions as a tumor suppressor, and its expression is lost in multiple types of human cancers (7,8).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
