Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human GATM protein.

Background

Mitochondrial glycine amidinotransferase (GATM), also known as arginine:glycine amidinotransferase (AGAT), catalyzes the conversion of glycine and L-arginine to guanidinoacetate and L-ornithine, the rate-limiting step of creatine biosynthesis (1-3). Mouse model studies of obesity-accelerated breast cancer show that this enzyme and creatine biosynthesis in adipocytes are essential to breast cancer progression driven by obesity (2). In addition, de novo synthesis of creatine by GATM or dietary uptake of creatine promotes tumor metastasis in an orthotopic mouse model (3). Moreover, GATM expression increases in metastases of various cancers, such as colorectal cancer and melanoma (3). Furthermore, creatine plays a role in neurons and its function requires GATM and SLC6A8, a creatine transporter (4).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
