Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

DEGS1 (F7G2J) Rabbit Monoclonal Antibody recognizes endogenous levels of total DEGS1 protein. This antibody does not cross-react with DEGS2 protein. This antibody may detect a nonspecific signal around 22 kDa.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human DEGS1 protein.

Background

Sphingolipid delta(4)-desaturase (DES1, DEGS1), also known as dihydroceramide desaturase-1, inserts a double bond into the sphingoid backbone of ceramides in most tissues. Studies show that deletion of Degs1 from ob/ob mice deficient in leptin improves glucose homeostasis and resolves insulin resistance and hepatic steatosis. These findings indicate that ceramides contribute to metabolic disorders (1). In addition, DEGS1 is involved in hematopoietic stem cell (HSC) function. In ex vivo culture, before quiescence exit, inhibition of DEGS1 in human HSCs by its inhibitor 4HPR activates stress response pathways, including the unfolded protein response (UPR) and autophagy, to maintain HSC self-renewal (2). Furthermore, DEGS1 is in the mitochondria-associated endoplasmic reticulum membrane (MAM) and is critical to MAM functions (3).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
