Product Usage Information

For optimal ChIP results, use 5 μL of antibody and 10 μg of chromatin (approximately 4 × 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.

Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

MTA3 (E3X2E) Rabbit Monoclonal Antibody recognizes endogenous levels of total MTA3 protein. This antibody does not recognize MTA1 or MTA2 protein. This antibody detects a 200 kDa protein of unknown origin.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val438 of human MTA3 protein.

Background

Metastasis associated gene 3 (MTA3) is a cell-type specific subunit of the NuRD deacetylase complex. Much like MTA1 and MTA2, MTA3 does not associate with other family members, forming a unique NuRD complex. MTA3 has been shown to be estrogen-dependent and a key regulator of Snail and E-cadherin in breast cancer (1). B lymphocyte differentiation has been shown to be dependent on the ability of BCL6 to use MTA3 as a cofactor (2). MTA3 has also been shown to repress the Wnt4 pathway (3). MTA3 activity has been described in many different cancer types, including breast, non-small cell lung cancer, and B-cell lymphomas (1,4,5).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Immunoprecipitation of MTA3 protein from HeLa cell extracts. Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is MTA3 (E3X2E) Rabbit mAb. Western blot analysis was performed using MTA3 (E3X2E) Rabbit mAb. Mouse Anti-Rabbit IgG (Light-Chain Specific) (D4W3E) mAb (HRP Conjugate) #93702 was used as a secondary antibody.

Chromatin immunoprecipitations were performed with cross-linked chromatin from K-562 cells and either MTA3 (E3X2E) Rabbit mAb or Normal Rabbit IgG #2729, using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR, using human Myc exon 2 primers and SimpleChIP® Human α Satellite Repeat Primers #4486. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the total amount of input chromatin, which is equivalent to one.
