Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

PPP1R17 (F3O5C) Rabbit Monoclonal Antibody recognizes endogenous levels of total PPP1R17 protein. Staining of uncertain specificity was observed in adipose tissue, intercalated discs of the heart, and kidney proximal tubules by immunohistochemistry.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of mouse PPP1R17 protein.

Background

Protein phosphatase 1 regulatory subunit 17 (PPP1R17) inhibits phosphatase activities of protein phosphatase type 1 (PP1) and protein phosphatase type 2A (PP2A) complexes (1). Its expression in the cerebellum is restricted to Purkinje cells, where it can be used as a marker for this cerebellar cell type (2). PPP1R17 expression has also been found in a subpopulation of neurons in the dorsomedial hypothalamus (DMH) that regulate restriction of food intake, aging, and longevity in mice (3,4).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
