Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA, 50% glycerol and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

INF2 (E7H5D) Rabbit Monoclonal Antibody recognizes endogenous levels of total INF2 protein. INF2 isoforms 1 and 2 appear as a doublet in the 170 kDa range using western blot analysis. This antibody also detects a band at approximately 100 kDa of unknown origin. This band may be a carboxy-terminal cleavage product of INF2.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro1019 of human INF2 protein.

Background

Inverted formin 2 (INF2) is a member of the formin family of actin assembly factors. INF2 regulates multiple cellular processes, including mitochondrial fission and vesicle trafficking. INF2 also regulates microtubule stability in conjunction with the formin mDia1 and IQGAP1 (1).
Mutations in INF2 have been linked to kidney disease and neuropathy in humans (2). INF2 stimulates Drp1-mediated and Drp1-independent mitochondrial division (3). Speckle-type POZ Protein (SPOP), a potential driver in human prostate cancer, regulates INF2 activity through ubiquitination, affecting INF2-mediated migration, invasion, and mitochondrial fission in prostate cancer (4). In thyroid cancer, JNK2 signaling mediates INF2-related mitochondrial fission to promote apoptosis (5).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
