    Background

    SH3-domain GRB2 Interacting Protein 1 (SGIP1) is an evolutionarily conserved protein primarily located in the brain. It is found in high concentrations near presynaptic nerve terminals, making up a significant 0.4% of the protein in those specific regions (1). The most established function of SGIP1 is its role in clathrin-mediated endocytosis (CME). For example, it functions as an endocytic adaptor for synaptotagmin 1 internalization at synapses (2). Further genetic variation in SGIP1 is associated with energy balance disturbances in humans, leading to obesity (3). More recently, SGIP1 has been associated with regulating mood and pain by affecting cannabinoid signaling (4). Loss-of-function (LOF) of the SGIP protein can lead to dysregulation of degradative organelles at the synapses and has been linked to Parkinsonism (5).
