Product Usage Information

For optimal ChIP results, use 2.5 μL of antibody and 10 μg of chromatin (approximately 4 × 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.

Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

MTF2 (F2X1A) Rabbit Monoclonal Antibody recognizes endogenous levels of total MTF2 protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly521 of human MTF2 protein.

Background

Metal regulatory transcription factor 2 (MTF2), also referred to as polycomb-like 2 (PCL2), is a catalytically inactive member of the PCL protein family (1). MTF2 binds to the metal response element of the mouse metallothionein promoter (2,3) and has also been shown to regulate the PRC2 activity at specific developmental genes in embryonic stem cells (4,5). It was also demonstrated that MTF2 promotes the proliferation and invasion of hepatocellular carcinoma cells (6) and that the MTF2–MDM2 axis can be targeted to sensitize refractory acute myeloid leukemia (AML) to chemotherapy (7).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Immunoprecipitation of MTF2 protein from mES cell extracts. Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is MTF2 (F2X1A) Rabbit mAb. Chromatin immunoprecipitations were performed with cross-linked chromatin from NCCIT cells and either MTF2 (F2X1A) Rabbit mAb or Normal Rabbit IgG #2729, using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR, using SimpleChIP® Human HoxA1 Intron 1 Primers #7707, SimpleChIP® Human HoxA2 Promoter Primers #5517, and SimpleChIP® Human α Satellite Repeat Primers #4486.
