Background

Zinc finger protein 281 (ZNF281), also known as ZFP281, ZBP-99, and ZNP-99, is a zinc finger-containing transcriptional factor. The protein binds to GC-rich sequences located in the regulatory regions of many genes to promote or repress target gene transcription (1). ZNF281 is involved in regulating many important biological processes. It promotes epithelial-mesenchymal transition by transcriptional upregulation of Snail (2), represses embryonic stem cell (ESC) pluripotency by down-regulation of Nanog transcription, an essential gene for cellular stemness (3,4), and orchestrates interconversion of pluripotent states of ESC by engaging expression of multiple genes such as EHMT1 and Zic2, and DNMT3 (5,6). DNA stability can be affected by ZNF281 in various ways. The protein facilitates DNA damage responses by controlling the expression of XRCC2 and XRCC4 (7), interacts with BRCA2 to prevent R-loop accumulation during DNA replication (8), and binds DNA breaks to facilitate DNA repair by non-homologous end joining (9). ZNF281 is upregulated and promotes tumor progression in most cancer types, such as colon cancer, pancreatic cancer, and breast cancer (10). ZFP281 locks early disseminated breast cancer cells in a dormant state and prevents cancer cell outgrowth in the lung metastatic niche (11).

Chromatin immunoprecipitations were performed with cross-linked chromatin from WERI-Rb-1 cells and either ZNF281 (F5Z7H) Rabbit mAb or Normal Rabbit IgG #2729, using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR, using human ZEB1 promoter primers and human CNTNAP4 downstream primers. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the total amount of input chromatin, which is equivalent to one.
