    Background

    EGF-like repeats and discoidin I-like domain 3 (EDIL3), also known as developmental endothelial locus-1 (DEL1), is a secreted protein that can interact with integrins (1,2). EDIL3/DEL1 is an anti-inflammatory protein that can be produced by endothelial cells, macrophages, neurons, and osteoclasts (3). EDIL3/DEL1, produced by endothelial cells, counteracts inflammation by limiting leukocyte recruitment. It binds to β2 integrins expressed on neutrophils, preventing them from interacting with ICAM-1 and disrupting neutrophil migration along the endothelium (3,4). EDIL3/DEL1 can also promote resolution by promoting macrophage clearance of apoptotic neutrophils, or efferocytosis, by acting as a bridge, binding macrophage αvβ3 integrin and apoptotic neutrophil phosphatidylserine (1,3). EDIL3/DEL1 deficiency is associated with several inflammatory conditions, including ankylosing spondylitis and multiple sclerosis (5,6). In cancer, however, EDIL3/DEL1 expression is often upregulated, promoting angiogenesis and epithelial-mesenchymal transition, making it a potential therapeutic target (7-9).
