Specificity/Sensitivity

MAZ (F3I2I) Rabbit Monoclonal Antibody recognizes endogenous levels of total MAZ protein. This antibody detects isoforms 1, 3, and 4 of MAZ protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human MAZ protein.

Background

The Myc-associated zinc finger protein (MAZ) is a transcription factor (TF) with four annotated isoforms and is widely expressed in most human tissues. This protein was independently identified as a regulator of MYC gene expression (1), activator of an insulin promoter (2), and modulator of serum amyloid A (3). Previous studies have shown that MAZ plays important roles in regulation of a wide variety of genes (4-6). Specifically, MAZ plays a dual role in regulating transcription initiation and termination processes. More recently, genome-wide screens have identified MAZ as a requirement for CTCF-mediated insulation (7). MAZ has also been shown to be highly expressed in malignant tumors and to promote tumorigenesis by directly binding to target gene promoters and cooperating with other TFs (8-13).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Simple WesternTM analysis of lysates (1.0 mg/mL) from HeLa cells using MAZ (F3I2I) Rabbit mAb #90940. The virtual lane view (left) shows the target band (as indicated) at 1:10 and 1:50 dilutions of primary antibody. The corresponding electropherogram view (right) plots chemiluminescence by molecular weight along the capillary at 1:10 (blue line) and 1:50 (green line) dilutions of primary antibody. This experiment was performed under reducing conditions on the JessTM Simple Western instrument from ProteinSimple, a BioTechne brand, using the 12-230 kDa separation module.
