Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

SCOT1/OXCT1 (F1O1D) Rabbit Monoclonal Antibody recognizes endogenous levels of total SCOT1/OXCT1 protein. This antibody does not cross-react with SCOT2/OXCT2 protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val180 of human SCOT1/OXCT1 protein.

Background

Mitochondrial succinyl-CoA:3-ketoacid CoA transferase 1 (SCOT1), also known as 3-oxoacid CoA-transferase 1 (OXCT1), catalyzes the rate-limiting step of ketone body oxidation by transferring CoA from succinyl-CoA to acetoacetate to form acetoacetyl-CoA (1,2). Studies show that SCOT1/OXCT1 expression and activity in skeletal muscle increase in obesity. Obesity-induced hyperglycemia is alleviated in skeletal muscle-specific SCOT1/OXCT1 knockout mice (1). In addition, SCOT1/OXCT1 functions as a lysine succinyltransferase. Succinylation of serine beta-lactamase-like protein (LACTB) by SCOT1/OXCT1 at K284 suppresses LACTB proteolytic activity, leading to hepatocellular carcinoma (HCC) progression (2). Furthermore, studies show that SCOT1/OXCT1 succinylation and activation mediated by SUCLA2 promote ketolysis and HCC cell proliferation in mice (3).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
