Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

Cystatin-A (F5N5B) Rabbit Monoclonal Antibody recognizes endogenous levels of total cystatin-A protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the amino terminus of human cystatin-A protein.

Background

Cystatin-A (CST-A), also known as cystatin SN or cystatin-1, is a member of the cystatin superfamily of proteins. Its primary function is the reversible and competitive inhibition of cysteine proteases, particularly lysosomal cathepsins (e.g., cathepsin B, H, and L). This inhibitory action is crucial for regulating cellular protein turnover, tissue remodeling, and maintaining cellular homeostasis (1). Our understanding of the role that cystatin-A plays in disease is incomplete. Cystatin-A is reported to be upregulated in several solid tumors, and elevated expression has been associated with enhanced cell proliferation, migration, invasion, and poor prognosis (2-4), suggesting that it might act as an oncogenic driver. However, some studies reporting decreased cystatin-A expression suggest it may play a protective role in certain tumor types (5,6).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
