Specificity/Sensitivity

TREM1 (D7D6I) Rabbit Monoclonal Antibody (Flow Specific) recognizes endogenous levels of total TREM1 protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the extracellular domain of human TREM1 protein.

Background

Triggering receptor expressed on myeloid cells 1 (TREM1) is a cell surface receptor found on neutrophils, monocytes, and macrophages, where it plays a critical role in regulating immune responses (1). TREM1 is activated by various extracellular ligands, including PGLYRP1, HMGB1, and eCIRP (CIRBP) (2). Upon activation, TREM1 associates with and induces the phosphorylation of its adaptor protein, DAP12, triggering downstream signaling pathways that drive proinflammatory cytokine production (3).Acting as an inflammatory amplifier, TREM-1 contributes to the pathogenesis of infectious diseases (4), non-infectious tissue damage (5), autoimmune disorders (6), and sepsis (7). In the central nervous system, TREM-1 expression in microglia promotes activation and polarization toward a proinflammatory phenotype, thereby facilitating disease progression (8). In the liver, TREM-1 is expressed on non-parenchymal cells (NPCs), where it stimulates the release of cytokines and chemokines and regulates lipid metabolism and fibrosis (9). Furthermore, TREM-1 promotes cancer cell proliferation, migration, and invasion by modulating tumor-associated inflammation and the microenvironment (10, 11). Consequently, targeting TREM-1 represents a promising therapeutic strategy for these diverse inflammatory conditions.

Flow cytometric analysis of human peripheral blood mononuclear cells, untreated (blue) or treated with LPS (300 ng/ml, 16 hr) #14011 (green), using TREM1 (D7D6I) Rabbit mAb (solid lines) or a concentration-matched Rabbit (DA1E) mAb IgG XP® Isotype control #3900 (dashed lines) co-stained with a CD14 antibody #36377. Anti-rabbit IgG (H+L), F(ab')2 Fragment (Alexa Fluor® 488 Conjugate) #4412 was used as a secondary antibody. Analysis was performed on CD14+ cells in the monocyte gate.
