Western blot analysis of extracts from NIH/3T3 cells, untreated or PDGF-stimulated (50 ng/ml for 5 minutes), using Phospho-PDGF Receptor beta (Tyr751) Antibody #3161 (upper) or PDGF receptor beta antibody (lower).
CST recommends using 50-100 ng/ml of PDGF for stimulation of PDGF signaling.
Platelet-Derived Growth Factor (PDGF) is supplied as a solution. It should be stored at –80°C. Aliquot the reagent upon receipt and avoid repeated freeze-thaw cycles.
The human PDGF-B coding cDNA was obtained from hepatocytic mRNA, subcloned into an expression vector and expressed in yeast. The recombinant human PDGF-BB dimer was purified and stored in PBS buffer containing 0.1% BSA.

Platelet-derived growth factor (PDGF) is a dimeric molecule that exists in homodimers or heterodimers of related polypeptide chains (A and B). Two types of PDGF receptors have been identified. The PDGF alpha-receptor binds all three PDGF isoforms with high affinity, whereas the beta-receptor binds only PDGF-BB with high affinity, PDGF-AB with low affinity and does not appear to bind PDGF-AA (1). Ligand binding induces receptor dimerization and autophosphorylation, allowing binding and activation of cytoplasmic SH2-domain-containing signal transduction molecules. A number of different signaling pathways are then initiated, leading to cell growth, actin reorganization, migration and differentiation (2-4). In clinical studies, PDGF expression has been demonstrated in a number of different solid tumors, from glioblastomas to prostate carcinomas. In these various tumor types, the biological role of PDGF signaling can vary from autocrine stimulation of cancer cell growth to more subtle paracrine interactions involving adjacent stroma and even angiogenesis. Targeting PDGF signaling is becoming an effective way for tumor treatment (5).

Any use of Product for diagnostic, prophylactic or therapeutic purposes, or any purchase of Product for resale (alone or as a component) or other commercial purpose, requires a separate license from CST. Customer shall (a) not sell, license, loan, donate or otherwise transfer or make available any Product to any third party, whether alone or in combination with other materials, or use the Products to manufacture any commercial products, (b) not copy, modify, reverse engineer, decompile, disassemble or otherwise attempt to discover the underlying structure or technology of the Products, or use the Products for the purpose of developing any products or services that would compete with CST's products or services, (c) not alter or remove from the Products any trademarks, trade names, logos, patent or copyright notices or markings, (d) use the Products solely in accordance with CST's Product Terms of Sale and any applicable documentation, and (e) comply with any license, terms of service or similar agreement with respect to any third party products or services used by Customer in connection with the Products.
