Description
The PathScan® TDP43 Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of TDP43. A TDP43 rabbit antibody has been coated onto the microwells. After incubation with cell lysates, TDP43 protein is captured by the coated antibody. Following extensive washing, a biotinylated TDP43 rabbit detection antibody is added to detect the captured TDP43 protein. HRP-linked streptavidin is then used to recognize the bound detection antibody. HRP substrate, TMB, is added to develop color. The magnitude of the absorbance for this developed color is proportional to the quantity of TDP43.

Specificity/Sensitivity
Background
TDP43 (TAR DNA-binding protein 43) is involved in transcriptional regulation and exon splicing (1,2). While normal TDP43 is a nuclear protein, pathological TDP43 is a component of insoluble aggregates in patients with frontotemporal lobar degeneration (FTLD) and amyotrophic lateral sclerosis (ALS). In these disorders, TDP43 is abnormally ubiquitinated, phosphorylated, and cleaved to generate carboxy-terminal fragments that are sequestered as insoluble aggregates in neuronal nuclei, perikarya, and neurites (3,4). Additionally, TDP43 inhibits the expression of the HIV-1 gene and regulates CFTR gene splicing (1,5).
