    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human AARS1 protein.

    Background

    Alanyl-tRNA synthetase 1 (AARS1) catalyzes the ligation of L-alanine to tRNA (1). It was later identified as a bona fide lactyltransferase that catalyzes protein lactylation using lactate and ATP. AARS1 senses lactate in the cell and activates the YAP-TEAD complex via lysine lactylation (1). In addition, studies show that AARS1 mediates H3K18 and STAT1 lysine lactylation to regulate fatty acid elongase-5 (ELOVL5) transcription, promoting ferroptosis in diabetic nephropathy (2). Furthermore, AARS1 was found to mediate global lysine lactylation in tumor cells, including that of p53. AARS1 expression is increased in many tumor types, correlating with poor prognosis among cancer patients with wild-type p53 (3). AARS1, along with AARS2 and its E. coli ortholog AlaRS, is considered a pan-lactyltransferase (4).
