Specificity / Sensitivity

Adora2b/Adenosine Receptor A2b (E8V7W) Rabbit mAb recognizes endogenous levels of total mouse Adora2b/Adenosine Receptor A2b protein. This antibody cross-reacts with human Adenosine receptor A2a protein but not with mouse Adenosine receptor A2a protein or human Adenosine receptor A2b protein. This antibody detects a 140 kDa band of unknown origin, and in human cell lines, detects a 42 kDa band of unknown origin.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of mouse Adora2b/Adenosine Receptor A2b protein.

Background

Adenosine receptor A2b (A2B adenosine receptor, A2BAR, ADORA2B) belongs to the adenosine receptor group of G protein-coupled receptors (GPCRs) (1-6). The adenosine receptors are divided into A1, A2, and A3 subtypes based on their ability to inhibit or stimulate adenyl cyclase (1-3). Within the A2 subtype, the high-affinity receptor is designated the A2A adenosine receptor, and the low-affinity receptor is the A2B adenosine receptor (1-3). Adenosine receptor A2b has been identified as a regulator of inflammation and vascular adhesion, primarily via signals from hematopoietic cells to the vasculature (3). Adenosine receptor A2b couples to G proteins to activate adenyl cyclase, leading to increased intracellular cAMP levels, as well as activation of phospholipase C (PLC), leading to increased intracellular calcium (4-6). Multiple mediators associated with inflammation, such as LPS, TNF-α, and IFN-γ, have been demonstrated to increase A2BAR expression in certain cell types (4-6).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
