Western blot analysis of extracts from SIM-A9 and C2C12 mouse cells, and mouse spleen using CD11b/ITGAM (E6E1M) Rabbit mAb (upper) and GAPDH (D16H11) XP® Rabbit mAb #5174 (lower). Data were generated using the standard formulation of this product.
Confocal immunofluorescent analysis of brain from the 5XFAD mouse model of Alzheimer’s disease using CD11b/ITGAM (E6E1M) Rabbit mAb (red). After blocking free secondary antibody binding sites with Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, the tissue was then labeled using β-Amyloid (D54D2) XP® Rabbit mAb (Alexa Fluor® 647 Conjugate) #42284 (green pseudocolor). Samples were mounted in ProLong® Gold Antifade Reagent with DAPI #8961 (blue). Data were generated using the standard formulation of this product.

Product Usage Information

This product is the carrier free version of product #17800. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

Formulation

Supplied in 1X PBS, BSA and Azide Free.

For standard formulation of this product see product #17800

Storage

Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

Specificity / Sensitivity

CD11b/ITGAM (E6E1M) Rabbit mAb (BSA and Azide Free) recognizes endogenous levels of total CD11b/ITGAM protein. Nonspecific signal is seen by western blot when testing human spleen tissue extracts.

Species Reactivity:

Mouse

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the amino terminus of mouse CD11b/ITGAM protein.

Background

Cluster of differentiation molecule 11b (CD11b)/Integrin alpha M (ITGAM) is a transmembrane protein forming heterodimers that are composed of α and β subunits (1). CD11b is expressed by, and commonly used as a marker for, myeloid lineage cells, including neutrophils, monocytes, macrophages, and microglia (2). CD11b is phosphorylated at Ser1126 (cytoplasmic tail) in neutrophils. Research has shown that this phosphorylation event plays a role for leukocytes traveling from the bloodstream to tissues (3). Furthermore, genome-wide association studies have linked CD11b to autoimmune diseases, such as systemic lupus erythematosus (SLE) (4).
