Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro12 of human CDK2AP1 protein.

Background

Cyclin-dependent kinase 2-associated protein 1 (CDK2AP1) was first identified as a tumor suppressor in oral cancer and is a member of the nucleosome remodeling and histone deacetylase (NuRD) complex (1-4). Also known as p12 or DOC-1, CDK2AP1 binds to unphosphorylated CDK2 and DNA polymerase alpha/primase to negatively regulate their activities (5,6). CDK2AP1 also plays a role in the silencing of Oct-4 during stem cell differentiation (7). Low CDK2AP1 expression in oral cancers is associated with poor prognosis (8). Lower expression has also shown to be resistant to cisplatin treatment (9). In addition to oral cancers, CDK2AP1 plays a role in other tumor types, including colorectal, testicular, and ovarian cancers (10,11).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

CDK2AP1 (E9D7P) Rabbit mAb

Western Blotting Image 1: CDK2AP1 (E9D7P) Rabbit mAb Expand Image
Western blot analysis of extracts from various cell lines using CDK2AP1 (E9D7P) Rabbit mAb (upper) or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower).
Immunoprecipitation Image 1: CDK2AP1 (E9D7P) Rabbit mAb Expand Image
Immunoprecipitation of CDK2AP1 protein from HeLa cell extracts. Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is CDK2AP1 (E9D7P) Rabbit mAb. Western blot analysis was performed using CDK2AP1 (E9D7P) Rabbit mAb. Anti-rabbit IgG, HRP-linked Antibody #7074 was used as a secondary antibody.
Chromatin Immunoprecipitation Image 1: CDK2AP1 (E9D7P) Rabbit mAb Expand Image
Chromatin immunoprecipitations were performed with cross-linked chromatin from K-562 cells, and either CDK2AP1 (E9D7P) Rabbit mAb or Normal Rabbit IgG #2729 using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR using human GAPDH exon 1 primers and SimpleChIP® Human Nanog Promoter Primers #95064. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the total amount of input chromatin, which is equivalent to one.
