    Background

    Class I-restricted T cell-associated molecule (CRTAM) is a type I transmembrane glycoprotein that plays a role in T cell development, proliferation, and adhesion (1). CRTAM is transiently expressed on the surface of activated CD8+ T cells and natural killer (NK) cells, where it binds to its ligand CADM1 (2,3). CRTAM is also expressed on particular subsets of activated CD4+ T cells and is required for the differentiation of CD4+ cytotoxic T cells (CTLs) (4-6). CRTAM binding to CADM1 has been shown to enhance the adhesive capacity and cytotoxicity of CD8+ T cells and NK cells (2). It also promotes retention and proliferation of CTLs in draining lymph nodes via interaction with CADM1-expressing dendritic cells (DCs) (7). Interestingly, elevated CRTAM expression has been found to be associated with increased survival in patients with triple negative breast cancer (TNBC), suggesting its use as a potential biomarker for immunotherapy efficacy (8).
