Product Information

Product Usage Information

For optimal ChIP results, use 10 μL of antibody and 10 μg of chromatin (approximately 4 × 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.
Application Dilution
Western Blotting 1:1000
Chromatin IP 1:50

Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

DGCR8 (E7A2J) Rabbit mAb recognizes endogenous levels of total DGCR8 protein.

Species Reactivity:

Human, Mouse, Rat, Monkey

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the carboxy terminus of mouse DGCR8 protein.

Background

Drosha was identified as a nuclear RNase III that catalyzes the initial step of microRNA (miRNA) processing (1). This enzyme processes the long primary transcript pri-miRNAs into stem-looped pre-miRNAs. Interference of Drosha results in the increase of pri-miRNAs and the decrease of pre-miRNAs (1). Drosha exists in a multiprotein complex called Microprocessor along with other components, such as DGCR8 (2). Drosha and DGCR8 are necessary for miRNA biogenesis (3).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Western Blotting Image 1: DGCR8 (E7A2J) Rabbit mAb Expand Image
Western blot analysis of extracts from 293T cells, transfected with control siRNA (-) or DGCR8 siRNA (+), using DGCR8 (E7A2J) Rabbit mAb (upper) or β-Actin (D6A8) Rabbit mAb #8457 (lower).

Western blot analysis of extracts from various cell lines using DGCR8 (E7A2J) Rabbit mAb.
Chromatin Immunoprecipitation Image 1: DGCR8 (E7A2J) Rabbit mAb Expand Image
Chromatin immunoprecipitations were performed with cross-linked chromatin from mES cells and either DGCR8 (E7A2J) Rabbit mAb or Normal Rabbit IgG #2729 using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR using mouse Nlrp12 exon 10 primers, SimpleChIP® Mouse Sox2 Exon1 Primers #30180, and SimpleChIP® Mouse MYT-1 Promoter Primers #8985. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the total amount of input chromatin, which is equivalent to one.
