Background

Rho family small GTPases act as molecular switches, regulating processes such as cell migration, adhesion, proliferation, and differentiation. They are activated by guanine nucleotide exchange factors (GEFs), which catalyze the exchange of bound GDP for GTP, and are inhibited by GTPase activating proteins (GAPs), which catalyze the hydrolysis of GTP to GDP (1). DOCK5 (dedicator of cytokinesis 5) is a Rho family GEF that plays an essential role in generating the sealing zone, an osteoclast-specific adhesion structure that mediates bone resorption and is regulated by actin and microtubule cytoskeletal signaling. Due to its role in bone resorption, DOCK5 is a potential therapeutic target for osteoporosis (2-4).

DOCK5 (E6R9W) Rabbit mAb

Western Blotting Image 1: DOCK5 (E6R9W) Rabbit mAb Expand Image
Western blot analysis of extracts from various cell lines using DOCK5 (E6R9W) Rabbit mAb (upper) or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower). Negative expression of DOCK5 protein in NCI-H446 cells is consistent with the predicted expression pattern.
Immunoprecipitation Image 1: DOCK5 (E6R9W) Rabbit mAb Expand Image
Immunoprecipitation of DOCK5 protein from DU 145 cell extracts. Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is DOCK5 (E6R9W) Rabbit mAb. Western blot analysis was performed using DOCK5 (E6R9W) Rabbit mAb. Anti-rabbit IgG, HRP-linked Antibody #7074 was used as a secondary antibody.
