Background

Found in inflammatory zone 1 (FIZZ1) protein, also known as resistin-like molecule α (RELMα) and hypoxia-induced mitogenic factor (HIMF), is a secreted protein with a cysteine-rich carboxyl-terminus (1). FIZZ1 is a member of the RELM family of proteins which consists of four isoforms in mice and two isoforms in humans. The murine family members are FIZZ1, FIZZ2, FIZZ3, and FIZZ4 and the human proteins are Resistin and RELMβ (2). FIZZ1 is expressed in lung, bone marrow, and spleen tissue (1,3). FIZZ1 transcription is mediated by a T helper type 2 (Th2) cytokine environment and its upregulation has been observed in inflammatory and infectious environments, including allergic airway inflammation, colitis, and helminth infection (4,5). FIZZ1 has been shown to drive myeloid cell chemotaxis and to downregulate Th2 cytokine production, both in a Bruton’s tyrosine kinase (BTK)-dependent manner (6,7). FIZZ1 is induced in a mouse chronic hypoxia model of pulmonary hypertension and activates the PI3K/Akt pathway, driving proliferation of pulmonary microvascular smooth muscle cells (8). FIZZ1 has been identified as a marker for alternatively activated macrophages, also known as M2 macrophages, which are characterized as anti-inflammatory and are hallmarks of several disease states, including infection, allergy, diabetes, and cancer (7,9,10).
