Western Blotting 1:1000
Chromatin IP 1:100

Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

HNF4γ (E4V2B) Rabbit mAb recognizes endogenous levels of total HNF4γ protein.

Species Reactivity:

Human, Rat, Monkey

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val363 of human HNF4γ protein.

Background

Hepatocyte nuclear factor 4γ (HNF4γ) is a transcription factor that belongs to the orphan nuclear receptor superfamily (1). Studies of Hnf4g knockout mice show that HNF4γ is involved in the regulation of glucose homeostasis (2,3). In addition, HNF4γ expression was shown to be upregulated in lung cancer. Overexpressed HNF4γ stimulates lung cancer cell proliferation (1). Furthermore, an aberrant gastrointestinal-lineage transcriptome was identified in about 30% of castration-resistant prostate cancers. HNF4γ, along with HNF1α, regulates this transcriptome. HNF4γ generates and maintains enhancers at the gastrointestinal-lineage genes as a pioneer factor and mediates castration resistance (4).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

HNF4γ (E4V2B) Rabbit mAb

Western Blotting Image 1: HNF4γ (E4V2B) Rabbit mAb Expand Image
Western blot analysis of extracts from various cell lines using HNF4γ (E4V2B) Rabbit mAb (upper) or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower). Negative expression of HNF4γ protein in BT-20 cells is consistent with the predicted expression pattern.
Western Blotting Image 2: HNF4γ (E4V2B) Rabbit mAb Expand Image
Western blot analysis of extracts from H-4-II-E and Vero cells using HNF4γ (E4V2B) Rabbit mAb.
Chromatin Immunoprecipitation Image 1: HNF4γ (E4V2B) Rabbit mAb Expand Image
Chromatin immunoprecipitations were performed with cross-linked chromatin from 22Rv1 cells and either HNF4γ (E4V2B) Rabbit mAb or Normal Rabbit IgG #2729 using SimpleChIP® Plus Enzymatic Chromatin IP Kit (Magnetic Beads) #9005. The enriched DNA was quantified by real-time PCR using human HNF1A promoter primers, human MUC13 promoter primers, and SimpleChIP® Human α Satellite Repeat Primers #4486. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the total amount of input chromatin, which is equivalent to one.
