Species Reactivity:

Human

Source / Purification

Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the central sequence of human JAM-A protein.

Background

Junctional Adhesion Molecule-A/F11 Receptor (JAM-A/F11R) is a transmembrane glycoprotein belonging to the immunoglobulin superfamily. JAM-A regulates multiple cellular processes, including tight junction assembly, epithelial-mesenchymal transition (EMT), leukocyte migration, virus binding, platelet activation, and angiogenesis.  Aberrant expression of JAM-A is correlated with poor patient prognosis in several human cancers (1,2). In a mouse model of atherosclerosis, an antagonistic peptide that inhibits JAM-A-expressing platelets from interacting with inflamed endothelial cells reduces atherosclerotic plaque formation (3).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

JAM-A (E6Z7E) Rabbit mAb

Western Blotting Image 1: JAM-A (E6Z7E) Rabbit mAb Expand Image
Western blot analysis of extracts from 293T cells, mock transfected (-) or transfected with a construct expressing full-length human JAM-A (hJAM-A-Myc/DDK; +), using JAM-A (E6Z7E) Rabbit mAb (upper), Myc-Tag (71D10) Rabbit mAb #2278 (middle), or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower).
Western Blotting Image 2: JAM-A (E6Z7E) Rabbit mAb Expand Image
Western blot analyasis of extracts from Capan-2, HaCaT, and BJ cells using JAM-A (E6Z7E) Rabbit mAb (upper) or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower). Low expression of JAM-A protein in BJ cells is consistent with the predicted expression pattern.
Immunoprecipitation Image 1: JAM-A (E6Z7E) Rabbit mAb Expand Image
Immunoprecipitation of JAM-A protein from HaCaT cell extracts. Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is JAM-A (E6Z7E) Rabbit mAb. Western blot analysis was performed using JAM-A (E6Z7E) Rabbit mAb. Mouse Anti-rabbit IgG (Conformation Specific) (L27A9) mAb (HRP Conjugate) #5127 was used as a secondary antibody.
Immunofluorescence Image 1: JAM-A (E6Z7E) Rabbit mAb Expand Image
Confocal immunofluorescent analysis of HaCaT cells (left, positive) and BJ cells (right, negative) using JAM-A (E6Z7E) Rabbit mAb (green), COX IV (4D11-B3-E8) Mouse mAb #11967 (red), and DAPI #4083 (blue).
