Immunoprecipitation of CHD4 from COS-7 cell extracts using Rabbit (DA1E) mAb IgG XP® Isotype Control #3900 (lane 2) or CHD4 (D8B12) Rabbit mAb (lane 3). Lane 1 is 10% input. Western blot analysis was performed using CHD4 (D8B12) Rabbit mAb.
Confocal immunofluorescent analysis of HeLa cells using HDAC2 (3F3) Mouse mAb (green). Actin filaments were labeled using DY-554 Phalloidin (red).
Western blot analysis of extracts from various cell lines using MBD3 (N87) Antibody.
Immunohistochemical analysis of paraffin-embedded mouse 4T1A mouse syngeneic tumor using MTA1 (D40D1) XP® Rabbit mAb.
Confocal immunofluorescent analysis of HeLa cells using RBAP46 (V415) Antibody (green). Actin filaments were labeled with DY-554 phalloidin (red).
After the primary antibody is bound to the target protein, a complex with HRP-linked secondary antibody is formed. The LumiGLO® is added and emits light during enzyme catalyzed decomposition.

NuRD Complex Antibody Sampler Kit offers an economical means of detecting each target protein that composes the nucleosome remodeling and deacetylation complex (NuRD). The kit contains enough primary antibody to perform two western blot experiments with each primary antibody.
All antibodies contained in this kit detect endogenous levels of their respective target protein.
Polyclonal antibodies are produced by immunizing animals with a synthetic peptide and are purified by protein A and peptide affinity chromatography. Monoclonal antibodies are produced by immunizing animals with recombinant human proteins or synthetic peptides.
The transcriptional repressor NuRD (nucleosome remodeling and histone deacetylase) complex is composed of multiple subunits, which include CHD3 (also known as Mi2-α), CHD4 (also known as Mi2-β), HDAC1, HDAC2, RBAP48, RBAP46, MTA1, MTA2, MTA3, and MBD3 (1-6). Both CHD3 and CHD4 contain two PHD (plant homeodomain) zinc finger domains that bind directly to HDAC1 and HDAC2. HDAC1 and HDAC2 are highly homologous and are involved in histone deacetylation, chromatin remodeling and transcriptional repression, and are also found together in the mSin3A and MeCP1 complexes (7-9). RBAP46 and RBAP48 bind to the histone fold region of histone H4 and are believed to target the NuRD complex to its histone substrates (10). MBD3, a member of the methyl-CpG-binding domain protein family, and the metastasis associated genes MTA1, MTA2, and MTA3 were all found to be integral components of the NuRD complex (2,3,7,11).
