Specificity / Sensitivity

UBQLN4 (E7N4Q) Rabbit mAb recognizes endogenous levels of total UBQLN4 protein. This antibody cross-reacts with a 150 kDa protein of unknown identity in some lysates and a 95 kDa protein of unknown identity in some rat lysates.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Lys87 of human UBQLN4 protein.

Background

Ubiquilin 4 (UBQLN4), a member of the ubiquitin-like (UBL) protein family, mediates protein degradation by binding polyubiquitinated targets as well as the S5a subunit of the proteasome (1,2). UBQLN4 acts as a repressor of homologous recombination-mediated DNA double-strand break repair and drives non-homologous end joining when overexpressed (3). Its role in DNA double-strand break repair is dependent on Ser318, which is phosphorylated by ATM in response to DNA damage (3). UBQLN4 expression is upregulated in colorectal cancer, and high expression is associated with decreased disease-free survival in several other cancers, including neuroblastoma, melanoma, and ovarian cancer (3,4). UBQLN4 has also been shown to directly associate with LC3 and UBQLN1, recruiting UBQLN1 to autophagosomes (5).
