Background

Ubiquitin-like PHD and RING finger domain-containing protein 2 (UHRF2), also known as Np95/ICBP90-like RING finger protein (NIRF), is a multi-domain member of the UHRF family and a UHRF1 paralog. Like UHRF1, UHRF2 carries out a variety of biological functions through its multiple domains, including a ubiquitin-like (UBL) domain, tandem Tudor domain (TTD), plant homeodomain (PHD), SET and RING finger-associated (SRA) domain, and really interesting new gene (RING) finger domain (1). Both UHRF1 and UHRF2 localize primarily with highly methylated pericentromeric heterochromatin and specifically recognize H3K9me2/3 marks (2,3). However, although they share high structural and sequence homology, UHRF1 and UHRF2 do not always carry out redundant biological functions. For instance, both UHRF1 and UHRF2 associate with hemi-methylated DNA through their respective SRA domains and complex with DNMT1, but UHRF2 does not target DNMT1 to replication forks and therefore cannot compensate for methylation defects in UHRF1- mouse ES cells (3-5). UHRF1 and UHRF2 also exhibit opposing expression patterns in differentiating cells and various tissues, further suggesting separate functions (6,7). UHRF2 may function as a novel cell cycle regulator by interacting with key proteins such as cyclin A2, CDK2, p53, and PCNA (7,8). Aberrant UHRF2 expression has also been implicated in several cancers, including lung and colorectal cancer as well as hepatocellular carcinoma (9-11).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
